      Chaetomium thermophilum is an eukaryotic thermophilic fungus that holds great potential for broad biochemical and biotechnological applications. We have developed a tool box for its genetic manipulation rendering this organism amenable as resource for structural biology. Here, we elucidate these approaches as a foundation for affinity purification of thermostable macromolecular complexes, like nucleoporin complexes and pre-ribosomal particles. [ABSTRACT FROM AUTHOR]
