The role of rafinose to maintain chilled semen quality of Garut ram's (Ovis aries) to support Indonesian sheep germplasm conservation.

      Garut sheep is one of Indonesia's biodiversity which needs to be protected. There are many efforts to conserve it, one of which is increasing reproductive efficiency with artificial insemination (AI). The aim of the research was to study the effectivity of raffinose to maintain sperm quality that is preserved at 3-5 0C. The research was conducted in four groups; tris extender containing 20% egg yolk (TEY-20) (T0), TEY-20 + Rafinose 0.2 g/100 ml (T1), TEY-20 + Rafinose 0.4 g/100 ml (T2) and TEY-20 + Rafinose 0.6 g/100 ml (T3) respectively. The examination of sperm quality including percentage of spermatozoa motility (SM), spermatozoa viability (SV), sperm membrane integrity (SMI), and sperm acrosome intact (SAI) was conducted every day over four days. The research design used in this research was a randomized complete design with four treatments and six replications. The percentage of SM in T3 (49.00%) was significantly (p<0.05) higher than T0 (43.00%) but was not significantly different (p>0.05) than T2 (48.00%). Evaluation of SMI showed that T4 (72.30%) was significantly (p<0.05) higher than T0 (64.50%). Evaluation of SAI showed that T4 (69.30%) was significantly (p<0.05) higher than T0 (60.00%). A dose of 0.6 g/100 mL raffinose was optimal to improve the quality of semen. [ABSTRACT FROM AUTHOR]
