- Abstract:
In order to grasp the structure function, diversity and regional and sex differences of the bacterial flora of the cecum of the mountain wild boars(Sus scrofa)in Guizhou, this paper reveals the specificity of its bacteria and the interaction of the host, the cecal bacterial flora of 12 Guizhou wild boars was analyzed by 16S rRNA high-throughput sequencing technology. Results A total of 59, 246 valid sequences were obtained, and 1, 256 OTUs were obtained by clustering with 97% similarity after optimization. The results showed as follows:(1)The cecal bacterial community of wild boars was abundant, and there was no significant difference in the bacterial community diversity between regions and sex groups, but there were some differences in specific bacterial community composition. (2)Copolymers were obtained from 278 genera of 18 phyla, the core flora includes six phyla, such as Firmicutes, Bacteroidetes and Actinobacteria. The core flora includes 16 dominant genera, such as Ruminococcaceae_UCG-005, Christensenellaceae_R-7_group and Prevotellaceae_NK3B31_group. (3)LEfSe analysis showed that there were significant differences(P<0. 05)between Qiandongnan(QDN)and Qianbei(QB)groups, such as Ruminococcus, Lactobacillus and Alloprevotella. (4)PICRUSt gene predicted and analyzed that 44 metabolic pathways were enriched in cecum of wild boars, mainly carbohydrate metabolism pathway, followed by amino acid metabolism pathway. In conclusion, the cecal bacterial flora diversity of Guizhou wild boars is higher than that of Henan wild boars, Chongqing captive wild boar and Tibetan pigs, and the core bacterial flora is enriched in different regions and between genders. The results of the study were to explore how the intestinal flora of the wild boars had the basic information of the interaction of the host, disease prevention and immunization development, and thus promote the study of the physiological and biochemical, protection management and population ecology of the wild boars. 为掌握贵州山地野猪(Sus scrofa)盲肠细菌菌群的结构功能、多样性以及菌群的地区和性别差异,揭示细菌的特异性及其与宿主的相互作用,采用16S rRNA高通量测序技术对12头贵州野猪盲肠细菌菌群进行分析,共获得有效序列59 246条,经优化后以97%相似度聚类得到1 256个OTU。结果表明:(1)不同地区和性别组间菌群多样性总体上差异均不显著,而在具体菌群组成上存在一定的差异菌属。(2)共聚类得到细菌18门278属,优势菌门为厚壁菌门(Firmicutes)、拟杆菌门(Bacteroidetes)和放线菌门(Actinobacteria)等6个,优势菌属有Ruminococcaceae_UCG-005、Christensenellaceae_R-7_group和Prevotellaceae_NK3B31_group等16个。(3)LEfSe分析表明,黔东南与黔北组间存在显著差异(P<0.05)的菌群为瘤胃球菌属(Ruminococcus)、乳杆菌属(Lactobacillus)和拟普雷沃氏菌属(Alloprevotella)等。(4)PICRUSt基因预测分析表明,野猪盲肠富集了44条代谢途径,以碳水化合物代谢途径为主,其次为氨基酸代谢途径。结合已有研究分析,贵州野猪盲肠细菌菌群多样性高于河南野猪、重庆圈养野猪及藏猪,不同地区、性别间均各自富集其核心菌群。研究结果为探究野猪肠道菌群如何与宿主相互作用、疾病预防和免疫发育提供基础资料,从而促进野猪的生理生化、保护管理和种群生态学等方面的研究。 [ABSTRACT FROM AUTHOR]
