- Abstract:
Single ascospore isolations were obtained from ascocarps of Y AASMYPL6 and Zhao00Ol of Morchella importuna, Y AASMJN5 of M. sextelata and Mes-19 Y AASM43 by capillary-isolation. These single ascospore isolat ions were analyzed for their mating type genes. The results showed that the four singleascospore populations contained either MAT] 1 1 or MAT] 2 1 with a ratio of about 1 : 1. A small number of single ascospore isolations of Zhao000l, Y AASMJN5 and Y AASM43 had both MATl 1 1 and MATl 2 1, and homokaryons with only one mating type gene were obtained from these isolations by mycelium-tip subculture, suggesting that some ascospores in Morchella were heterokaryotic. Paired-cultivation for singleascospore isolation populations of Y AASMYPL6, Zhao00Ol and Y AASM43 showed that there was very few vegetative compatibility in MATl 1 X MATl 1, MATl 2 X MATl 2 or MATl 1 X MATl 2. Whether the phenomenon that two strains are sexual compatible but at the same time vegetative incompatible exists in crossbreeding of basidiomycete fungi is st ill awaiting more attentions. 以人工栽培梯校羊肚菌(Morchella importuna)千囊果 YAASMYPL6 和六妹羊肚菌(M. sextelata)子 囊果YAASMJN5.野生梯校羊肚菌子果 Zhiyi001 和黃色羊肚菌(Mes-19)子囊果YAASM43为材料,不用 毛细管单孢分离法分离其子囊抱子获得单孢菌株.并通过基于交配型基因的分子标记方法鉴定单在菌株的交 配型,结果显示14个样本群体中其有交配型基因 MAT1-1-1和 MAT 12-1的单孢菌株比例为1:1.3个子囊果 的单胞菌株群体中检测到同时具有MATI-1-1和MAT1-21的单型菌株,且这样的营体经过多次失端菌丝传 代培养后可获得仅有一种交配型基因的同核体商株,证明单抱菌株群体中存在一定比率的异核体的株,在 YAASMYPL6.Zhao0001和YAASM43 3个样本群体内选择具有相同和不同交配型基因的 单孢菌株进行对 時培养。发现对蜂组合中表现营养体和的组合数量极少,性亲和但因营养体不亲和面表现出菌丝指抗的现 象。在已知食用担子菌的杂交有种中是否存在尚未被关注.
