Con el objetivo de caracterizar morfométricamente los ovocitos de Bubalus bubalis, se evaluaron 274 complejos cúmulos-ovocitos, extraídos por el método de disección quirúrgica de los folículos superficiales de 48 ovarios. Los ovocitos se obtuvieron mediante la maduración in vitro, La captura de los mismos se realizó bajo visión estereoscópica con capilares y se procedió a removerlos de las células del cúmulo. Los indicadores evaluados fueron: diámetro total del ovocito, diámetro del citoplasma y espesor de la zona pelúcida. Los ovocitos se maduraron en TCM 199 con 10% de SFB durante 24 h a 37oC. A las 24 horas de maduración in vitro se les realizaron las mismas mediciones bajo visión microscópica. Los datos se procesaron por prueba T para comparación de medias. Los valores obtenidos fueron para el diámetro total 154,61µm vs. 167,14µm, para el citoplasma 145,21µm vs. 148,46µm y para la zona pelúcida 7,85µm vs. 17,79µm (ovocitos inmaduros vs. maduros, respectivamente). Se evidenció que el incremento del diámetro total responde fundamentalmente al aumento del grosor de la zona pelúcida en 9,94 µm. Este tipo de caracterización se realiza por primera vez en Cuba para Bubalus bubalis y es parte de los elementos previos para la realización de biotecnologías relacionadas con el proceso reproductor. In order to morphometrically characterize Bubalus bubalis oocytes, 274 oocyte-cumulus complexes were evaluated. They were extracted by the surgical dissection method of the superficial follicles of 48 ovaries. The oocytes were obtained by in vitro maturation. Their capture was carried out under stereoscopic vision with hair, removing the cumulus cells. The indicators evaluated were: total diameter of oocyte, cytoplasm diameter and thickness of the pellucid zone. Oocytes were matured in TCM 199 with 10% FBS for 24 hours at 37oC. After 24 hours of in vitro maturation, the same measurements under microscopic vision were performed. Data were processed by t-test for mean comparison. The values obtained were: for the overall diameter: 154.61µm vs. 167.14µm, for cytoplasm: 145.21µm vs. 148.46µm and for the pellucid zone: 7.85µm vs. 17.79µm (immature oocytes vs. mature oocytes, respectively). It was evidenced that the overall diameter increase responds primarily to the thickness increase of the pellucid zone (PZ) at 9.94µm. This type of characterization has been carried out for Bubalus bubalis for the first time in Cuba, and it is part of the previous elements in performing biotechnologies related to the reproductive process.
